The stained sections were examined and digitized using a confocal microscope (Leica TCS-SP).
Previously it has been shown that a targeting vector, pMP8, can correct the loss of Hprt activity in E14TG2a ES cells that lack exons 1 and 2 of the hprt gene [ 2 15 ] . The design of this vector implied that it should also be capable of correcting the hprt exon 1 deficiency in F3 cells and could, therefore, be used to introduce transgenes into the hprt locus by homologous recombination.
