The GTPase can then re-enter the transport cycle through nucleotide exchange promoted by a specific exchange factor at the donor vesicle membrane [ 18 19 20 ] . In addition to changes in guanine nucleotide state, the posttranslational prenylation of Rab proteins plays an important role in the cycling mechanism.

  • The C-terminal 30 amino acids of Gα consist of the β6-α5 loop, followed by the α5 helix and finally several amino acids of flexible structure (Figure 1) [ 19 20 21 ] . The β6-α5 loop stabilizes the guanine ring of bound GDP or GTP, and mutations in this region affect guanine-nucleotide binding.

  • Guanine and cytosine gave intermediate values, guanine being in both cases higher than cytosine, although this difference is only significant in the CAN series.

  • An intuitive approach to the problem that examines sequence composition is to compare the guanine and cytosine (GC) base content of a sequence with other sequences from the species being studied.
