Our finding that KRC binds to the RSS as well as the κB motif may also provide a link between transcription and recombination in the context of the accessibility model [ 58 59 ] . Enhancer or promoter elements are important for the recombination process in cell lines and animal models [ 60 ] . Similarly, expression of transcription factors, in conjunction with the recombination activating genes, has been shown to induce V(D)J recombination in non-lymphoid tissues by rendering RSS accessible to the recombinase [ 61 ] . The κB motif, first found in the Igκ light chain [ 62 ] , and later in the TCR β2 locus [ 31 ] , has been shown to promote V(D)J recombination by modulating locus accessibility [ 63 ] . In addition to influencing recombination by binding of RSS, KRC binding of the κB motif may modulate accessibility and transcription of target loci.
During this transition, T cell precursors sequentially gain and lose cell surface expression of CD44 and CD25 markers, express the pre-T cell receptor (pTCR), and undergo β-selection [ 2 7 8 9 ] . The other transition is from CD4 +8 +DP to CD4 +or CD8 +single positive (SP) thymocytes, in which DP cells express CD3, rearrange their α-TCR, undergo positive and negative selection, and down regulate CD4 or CD8 to become functional CD8 +or CD4 +SP T cells [ 10 11 ] . In addition to functional and cell surface marker changes, each thymocyte differentiation stage is marked by characteristic changes in proliferation and apoptosis [ 12 13 14 15 16 17 18 ] .
Mounting evidence suggests that the pre-TCR does not require interaction with a ligand to perform its function, and the subsequent CD44 -25 -(TN4 or pre-DP) to DP transition has been termed "programmed differentiation" to indicate that this step can occur in isolated thymocytes, presumably without input from extrinsic signals [ 25 26 27 ] . However, there is also evidence that the thymic microenvironment does contribute signals required for the TN-DP transition, particularly to promote thymocyte proliferation and apoptosis [ 13 28 29 30 ] . The specific nature and purpose of the TEC contribution during this important transition therefore remains to be determined.
In agreement with previous observations, little infiltration was detected in Rip1-Tag2 mice receiving TCR splenocyte transfer (Figure 3).
Since only a proportion of developing thymocytes succeed in expressing the α:β TCR heterodimer, PKC activation also provides the signal to turning off the CD4 and CD8 genes on cells that failed to express the TCR, which are non-functional and potentially harmful.
