  • The initial step of the insertion can proceed in the presence of either Mg.ATP or a non-hydrolyzable ATP-analog and involves oligomerization of the BchD and BchI subunits to form an oligomeric ring of AAA protomers that resembles the ring structure of NSF and other AAA proteins [ 14 32 ] . The second step of the insertion involves an obligatory hydrolysis of ATP that is tightly coupled with the transfer of the chelated Mg 2+to the protoporphyrin ring by BchH [ 49 ] . Although the details differ from midasin, particularly with respect to the subunit composition of the AAA ring, this chelation reaction provides a structural model suggesting that the function of the midasin M-domain may be to regulate the ATPase activity of the AAA protomers in the pseudo-hexameric ring and thus couple ATP hydrolysis to the binding of a protein ligand at the MIDAS site.

  • Fas contains a single transmembrane domain and belongs to the tumor necrosis factor (TNF)/nerve growth factor family [ 8 ] . FasL contains a single transmembrane domain and is also a member of the same TNF family [ 9 ] . A soluble form of FasL has been described, but appears to be less capable of inducing apoptosis, when compared with the bound form [ 10 11 ] . The binding of FasL with Fas initiates receptor oligomerization, which recruits Fas-associated death domain (FADD) [ 12 ] . FADD binds procaspase-8 and permits activation of caspase-8 through self-cleavage [ 13 ] . Caspase-8 activates the effector caspases, which commits the cell to the orderly process of apoptosis [ 14 15 ] . In addition, caspase-8 cleaves Bcl-2-interactive-death-agonist (Bid) [ 16 ] . Truncated Bid localizes to the mitochondria and promotes cytochrome c release; this process also serves as a major apoptotic-signaling pathway for Fas [ 16 17 18 ] . Depending upon the amount of caspase-8 that is activated, the predominant pathway can be either Bid cleavage with subsequent mitochondrial release of cytochrome c or activation of the effector caspase pathway [ 18 ] . Resident cells of the kidney express both Fas and FasL and Fas/FasL signaling is functional in these cells [ 19 20 21 22 23 24 ] . A recent review by Ortiz and associates [ 25 ] noted that the combined literature demonstrating participation of Fas/FasL pathway in renal injury essentially fulfilled Koch's postulates.

  • Although the energy required for lateral separation of the transmembrane domains in membrane appears to be high, the third step in Figure 1 (clustering via transmembrane domain oligomerization) may compensate for it.

  • The N-terminal core region of high similarity between family members correlates to a region in nucleophosmin/B23 that has been shown to be involved in oligomerization as well as chaperone activity [ 44 ] . A central portion between nucleophosmin's two acidic domains is required for ribonuclease activity [ 44 ] ; this region does not have a corresponding domain in either NPM3 or nucleoplasmin, suggesting that these proteins would lack such activity.

  • SAM domain oligomerization is the key event that triggers a variety of leukemias [ 21 ] and is therefore an attractive target for therapeutic intervention.
