Our data in DMSO-water show the same features, which are expected from the complex kinetics of nucleation and growth of insoluble aggregates of UCB diacid (H 2 B), leading to the formation of a new solid phase [ 19 20 ] . Our centrifugation, 5 min at 14,000 g , was quite mild, and the short 20-minute period between preparation of UCB-DMSO-water systems and centrifugation severely limited the time-dependent growth to large aggregates.
One process inhibited by BDM is the centripetal flow of actin from the leading edge into the cell body, known as retrograde flow [ 11 12 ] . Retrograde flow is thought to be driven by a "push-pull" mechanism of polymerization of actin at the leading edge of the cell and contractile activity of myosin motor proteins [ 13 14 ] . Formation of new actin filaments is predominantly mediated by the nucleation activity of the Arp2/3 complex [ 15 ] . Members of the WASP/WAVE family of proteins, among others, stimulate and regulate the Arp2/3 complex for spatial and temporal control of filament nucleation [ 15 ] . Localization of these proteins to the leading edge, though not completely understood, is probably mediated through their interactions with the actin cytoskeleton itself and membrane-associated binding partners [ 15 16 ] .
During this transition, tightly-regulated post-translational phosphorylation-dephosphorylation events and proteasome-mediated proteolytic reactions regulate the activation and inactivation of signal transduction pathways that control chromatin condensation, nuclear membrane dissolution, microtubule nucleation, and formation of a haploid oocyte [ 8 9 10 ] . Several kinases that exert major roles during OM include maturation promoting factor (MPF) [ 9 11 ] , mitogen-activated protein kinases (MAPKs) [ 12 13 ] , and the product of the c-mos protooncogene Mos kinase [ 14 15 ] . Also, during OM oocytes undergo two highly-regulated metaphase-anaphase transitions (MAT) in which homologous chromosomes are equally and randomly segregated to an oocyte and first polar body and a subsequent division in which equational division of sister chromatids results in a haploid oocyte and a second polar body.
Since actin incorporation is not due to elongation of existing filaments and BDM does not affect pure actin dynamics in vitro, we examined the localization of Arp3, a subunit of the actin nucleation Arp2/3 complex.
Future studies will hopefully clarify the role of actin nucleation in the Aplysia system or alternative mechanisms that would reconcile the discrepancies between systems.
