Mammalian microtubules are formed from a mixture of α-tubulin and β-tubulin isotype classes [ 11 15 16 ] , and the antimitotics used in chemotherapy interact primarily with β-tubulin.

  • In ovarian cancer cells, mutations in residues near the putative paclitaxel-binding site of β-tubulin class I were found in paclitaxel-resistant ovarian cancer cells [ 23 ] . Other work suggests that changes in tubulin isotypes alone cannot confer resistance to antimitotics.

  • Common agents currently used in treating metastatic breast cancer are the antimitotics paclitaxel and docetaxel [ 1 2 ] . These drugs bind to β-tubulin, a major protein in mitotic spindles, and halt cell division at metaphase.
