Overexpression of eIF4E leads to deregulated growth and malignant transformation of a variety of cultured cell lines [ 12 13 14 ] . Moreover, elevated levels of eIF4E are commonly found in solid tumors, especially in breast, colon, and head and neck tumors [ 15 ] . Clinical studies indicate that eIF4E gene amplification and protein overexpression is associated with malignant progression in these tumors [ 16 ] . High eIF4E levels are also associated with a higher rate of cancer recurrence and cancer-related death [ 17 ] . In contrast to eIF4E, overexpression of 4EBP inhibits cell proliferation [ 18 ] and 4EBP-1 expression levels are inversely correlated with the progression of certain types of tumors [ 19 ] .
Chemoprevention has been defined as the use of noncytotoxic nutrients or pharmacologic agents to enhance intrinsic physiologic mechanisms that protect the organism against the development of mutant clones and their progression to malignant cancer [ 27].
A poor response of tumor cells to IFNg is in agreement with previous observations that many tumors acquire such resistance upon malignant transformation, possibly important as a mechanism of tumor escape from immune surveillance [ 29 ] . The nature of this phenomenon is complex, and multiple defects that can prevent IFNg responses in tumor lines have been described.
The retina and malignant solid tumors also produce the sulfated GAGs [ 1 2 22 23 24 25 26 ] . Although HSs are mostly found as proteoglycans associated with the basal lamina or the plasma membrane, the presence of HSs in the culturing medium might be a result of shedding or release of their extracellular domains from the cell membranes as soluble components.
Several lines of evidence indicate that NF-κB is constitutively activated in cancer cells, suggesting that it plays an important role in tumorigenesis [ 30 ] . Activated glucocorticoid receptors have been shown to repress NF-κB-mediated gene expression [ 31 ] , and to cause a significant reduction in nuclear levels of p65 protein, a subunit of NF-κB [ 31 ] . A variety of mitogens, cytokines, and reactive oxygen intermediates have been shown to rapidly cause the translocation of NF-κB to the nucleus [ 30 31 32 ] , and the ability of dexamethasone to interfere with this translocation is blocked by prior treatment with the glucocorticoid antagonist RU 486 [ 30 ] . Based on these findings, it is plausible to speculate that exposure to RU 486 may interfere with the function of endogenous glucocorticoids as regulators of cell cycle, allowing the initiation and progression of malignant transformation of liver cells.
