Several groups developed a more direct approach for determining occupancy by analyzing effects on the composition of the final complex, after in vitro reconstitution, when amino acid residues in LHCPs were replaced with substitutes that are unable to serve as a ligand.
Experiments with the model alga Chlamydomonas reinhardtii [ 11 ] showed that LHCPs were not detectably imported into the chloroplast in the absence of Chl synthesis and instead accumulated outside of the chloroplast in the cytosol and in vacuoles [ 12 ] .
