  • A high prevalence of 3q amplification has been reported in many studies on tobacco-associated cancers, including those of the lung, cervix and esophagus [ 11 13 14 15 16 17 ] . We and others have shown that PIK3CA is a candidate oncogene at 3q in these tumors [ 12 18 19 ] . Interestingly, PI3K pathway has been suggested to play a role in modulating the effects of benzopyrene, a tobacco-derived carcinogen, and its metabolite, anti-7,8-dihydroxy-9,10-epoxy-7,8,9,10-terahydrobenzopyrene [ 20 ] .

  • Some investigators are now recommending that lower PSA cutpoints should be used as an indication for prostate biopsy [ 37 38 39 ] . Catalona and colleagues detected cancer in 22% of men biopsied with PSA levels between 2.6 to 4 ng/ml [ 37 ] and Lodding and colleagues detected cancers in 13% of men with PSA values between 3 to 4 ng/ml [ 39 ] . We found that lowering the cutpoint to 2 ng/ml, while greatly increasing sensitivity, led to an 80% false positive rate.

  • Rare SPAM1 transcripts are however not ubiquitous as revealed by their absence from human ovary, spleen, and liver [ 21 ] and murine skeletal muscle [ 22 ] . Both rare and abundant SPAM1 transcripts have been found in neoplastic breast tissue [ 23 ] and in a number of other cancers including pharyngeal [ 24 ] , metastatic melanomas and gliomas [ 25 ] . In normal somatic cells rare transcripts have been found in breast tissue [ 23 ] and in fetal, placental, and prostate cDNA libraries [ 21 ] .
