The assembly of myosin II monomers into filaments is regulated by phosphorylation of its heavy chains at three threonine residues at the C-terminus of the tail [ 9 10 ] . Dephosphorylation of these threonines is a prerequisite of filament assembly, as confirmed by the phenotypes of a 3xAsp mutant, in which the three threonines are replaced by three aspartate residues (mimicking the phosphorylated state) [ 11 ] .
