The midasin gene in Giardia intestinalis (accession AF494287) was cloned in silico from sets 1-11 of the unassembled genomic nucleotide sequences kindly made available by the Giardia Genome Project [ 11 ] . Regions of conserved amino acid sequence in the human and yeast genes for midasin were used as input to the Tblastn server at Genbank [ 56 ] to identify clones that encoded homologous regions in Giardia.
The ortholog in these species, as well as the presence of an unassembled orthologous gene in humans, Caenorhabditis and Giardia, has been noted by Mocz and Gibbons [ 6 ] , who discussed its evolutionary relationship to the heavy chain of dynein motor protein.
We used the following genomic sequences and analyses in this work: D. melanogaster Release 3 sequence (Berkeley Drosophila Genome Project, BDGP) [ 36 ] ; D. melanogaster Release 3.1 annotation (BDGP) [ 40 ] ; D. pseudoobscura Release 1 sequence and assembly (Human Genome Sequencing Center at Baylor College of Medicine, HGSC at BCM) [ 37 ] ; whole-genome alignment of D. melanogaster and D. pseudoobscura (Berkeley Genome Pipeline) [ 32 ] ; Anopheles gambiae assembled genomic sequence (Anopheles Genome Project) [ 56 ] ; Apis mellifera unassembled genomic trace sequence (HGSC at BCM) [ 48 ] .
Unexpectedly, the Anopheles genome contains only four members of this subfamily, and all are located in a single cluster on 2L (Figure 7b); the same four members were identifiable in the unassembled genomic sequence of Apis [ 48 ] . The simplest scenario is that members of the K box-family have undergone radical duplication and dispersal about the genome in Drosophila . This is consistent with the finding that the remaining members of the K-box family [ 47 ] , including mir-11 , the three mir-6 paralogs and the K-box-antisense gene mir-5 , are similarly absent from both Anopheles and Apis . However, one new putative member of the K-box family (2R:4681879) was identified by miRseeker, and it has been conserved in all four sequenced insect genomes.
In Caenorhabditis, there appears to be an unassembled midasin gene located on chromosome VIII (data not shown).
