The possibility that domoic acid may activate neonatal microglia both in vitro and in vivo is suggested by recent observations with kainic acid, a domoic acid and glutamate analog that can induce upregulation of neonatal microglia scavenger receptor mRNA [ 22 ] and tissue plasminogen activator release [ 23 ] . The putative molecular explanation for these observations has been that kainic acid activates the ionotropic AMPA glutamate GluR4 subunit which has recently been shown to be expressed by activated microglia following transient forebrain ischemia in vivo [ 24 ] . Because this subunit of the AMPA glutamate receptor has affinity for domoic acid [ 25 26 27 ] , we hypothesized that domoic acid might activate neonatal microglia in vitro and lead to concomitant mediator release [ 21 ] .
Injured adult DRG neurons do not die after axonal injury [ 15 ] as a result of the upregulation of cell survival factors such as the small heat shock protein HSP27 [ 16 ] . We now find several genes that could have a survival-promoting role, including the peripheral benzodiazepine receptor, whose agonists are potent antiapoptotic compounds [ 17 ] , GADD45A, which maintains genomic stability and appears to be anti-apoptotic in neurons after ischemic damage [ 18 ] , metallothionein 1L, which is a free radical scavenger [ 19 ] , GTP cyclohydrolase 1, which is obligatory for the actions of NGF on PC12 cells [ 20 ] , and IES-JE (MCP-1) which attracts macrophages, which in turn have been reported to have a role in neuronal survival [ 21 ] .
Our finding that AGE-BSA and PA induce apoptosis through a CD36-mediated and p38- and caspase-dependent mechanism in tubular epithelial cells, similar to TSP-1 and ox-LDL in endothelial cells and macrophages, respectively, suggests that multiple, context-dependent extracellular stimuli of apoptosis may converge on CD36 scavenger receptor to activate src kinase and proapoptotic p38 MAPK pathway.
Branch 1 of the HIT superfamily includes the ubiquitous Hint enzymes [ 25] plus two enzymes with a more phylogenetically resstricted distribution, namely Aprataxin, which is lost in humans with ataxia-oculomotor apraxia [ 26, 27] and the scavenger mRNA decapping enzyme Dcps [ 28, 29].
It was a scavenger's revolution that held large promise for a time: finding potential in the most random litter, making a new world out of the debris of the old.
