Studies in budding yeast identified SCF Cdc4p, an E3 ubiquitin ligase complex that mediates the ubiquitylation of the CDK inhibitor Sic1p [ 3 4 ] . SCF Cdc4pconsists of at least four proteins: the cullin Cdc53p, the RING domain protein Hrt1p/Rbx1p/Roc1p, the adapter protein Skp1p, and Cdc4p (reviewed in [ 1 ] ). All human cullins interact with HRT1/RBX1/ROC1 [ 7], are modified by NEDD8 [ 8], and have ubiquitin ligase activity in vitro [ 9].
