  • It has been proposed that loss of hERα expression in breast tumors is the result of aberrant RNA processing [ 40 41 42 43 ] . These aberrantly spliced mRNAs, if translated, could lead to truncated receptors with hormone binding but not DNA binding function [ 44 ] . The ability of Nova-1 to activate exon selection in neurons is antagonized by a second RNA-binding protein, brPTB (brain-enriched polypyrimidine tract-binding protein), which interacts with Nova-1 and inhibits its function [ 24 ] . DAZAP1 could function in a similar manner by binding to DAZL and inhibiting its function.

  • Group II introns were first identified in fungal mitochondria and grouped on the basis of their secondary structures [ 2 4 5 ] . They have been found in organelle genomes of fungi and plants and also in a number of bacteria [ 6 ] . None have been found in animals, although it has been suggested that group II introns may be progenitors of nuclear spliceosomal introns based on similarities between nuclear pre-mRNA splicing in eukaryotes and the splicing mechanism used by group II introns [ 7 8 ] .

  • However, this possibility contradicts current models of exon definition in which exons in pre-mRNA are first defined by proteins interacting across exons and then processed as relatively autonomous units.

  • This finding is not anomalous because analysis of exon sequences surrounding the total set of introns in S. cerevisiae pre-mRNA genes shows a preference for AAAG at the 5' splice site [ 31 ] . The final G in this motif has been established as significantly conserved in yeast [ 32 ] . The sequence at the proximal 5' exon region is required for interactions with the spliceosomal small nuclear ribonucleoprotein particle U1 [ 19 ] . Our data are, therefore, consistent with present understanding of yeast pre-mRNA splicing.
