  • Repetitive proline sequences tend to adopt a polyproline II (PPII) helix, consisting of an extended structure with three residues per turn.

  • For example, the junction between the aptamer and the Group I ribozyme in the aptazyme Th2P6 resembled the deletion variant td ΔP6-6; the aptazyme was activated by theophylline, while the splicing defect of the deletion variant at 3 mM magnesium was suppressed by 8 mM magnesium or by stabilization of a tetraloop sequence that capped the P6 helix [ 15 ] . The pattern of proteolysis and sites of cleavage for Ppt1p are very similar to those seen with PP5 under these same conditions [ 19 ] . This indicates that Ppt1p also contains an accessible region linking the TPR and catalytic domains and is most consistent with the presence of three TPRs in Ppt1p.

  • The residues K10, the backbone of S11, V12, V12, S14, V17, G40, E55, K63 and 64V, from the kinked carboxy-terminal helix, a beta-turn, and parts of the beta-sheet, define a putative binding epitope (Figure 2b).

  • Although the α5 helix (Figure 1) does not directly contact the bound guanine-nucleotide [ 19 20 21 ] , it is clearly important for Gα structure and to maintain the proper orientation of the β6-α5 loop.
