  • Fiber might affect the risk of breast cancer by decreasing estrogen levels; fiber binds estrogen excreted into the gastrointestinal tract and reduces enterohepatic circulation [ 17 18 ] . In addition, higher dietary fiber intake has been associated with higher plasma levels of sex-hormone-binding globulin, which can reduce the bioavailability of estrogens [ 19 ] . The potential mechanism of a protective effect of vegetable fat is more speculative: vegetable fat is a composite of multiple forms of fatty acids, some of which have been proposed to increase risk (such as C 18:2 n-6) whereas other specific fatty acids have been proposed to decrease risk (such as C 18:1 , the primary fatty acid in olive oil) [ 20 ] . More detail on the type and composition of the vegetable oil used is necessary to pursue this relation with greater precision.

  • In fact, activation of TGF-β requires the binding of latent TGF-β to the M6P/IGF2 receptor [ 11 ] . Therefore, down-regulation of M6P/IGF2R expression could also lead to a decreased bioavailability of activated TGF-β, thereby decreasing the sensitivity of MCF-7 cells to the TGF-β apoptotic pathway.

  • Although we cannot eliminate the possibility of more subtle effects of chemopreventive agents on TGF-β bioavailability or cellular responsiveness, in our preliminary analyses we have seen no effects on the expression of type I and type II TGF-β receptors (data not shown).

  • Compounds III with X = O, Y = H and OH in position 1, and compound II with X = S and Y = H were selected as the most promising candidates since their predicted oral bioavailability was among the highest while the potential harmful side effects were predicted to be low (Table 4).

  • Thus, they were tested against databases predicting plasma protein binding affinity [ 32 ] , oral bioavailability and urine excretion (unpublished results).
