While we did not perform a detailed comparison of EGFP expression in FLS using the modified and unmodified vector backbones, preliminary experiments suggested that the modified vector was superior (J Yang, unpublished observations).

  • The importance of such bonds for activity was shown in other work, where huAPE1's binding to the DNA backbone is only inhibited by ethylation of the phosphates two and three positions 3' to an abasic site [ 18 ] . Mutation of R177A, at the end of Molego 6, that binds to this region and to the bases opposite the AP-site had enhanced activity [ 11 ] , while mutations at W280 (Molego 11) and F266 (Molego 10) [ 33 ] reduce activity and, in the latter case, substrate selectivity.

  • The adenovirus (AV) backbone for the aquaporin-1 (AQP1) sense and antisense AV constructs was a replication-deficient "first-generation" AV with deletions of the E1 and E3 genes [ 26 ] . This "empty" AV contains the cytomegalovirus (CMV) promoter and bovine growth hormone polyadenylation (bHG) site separated by a polylinker that was used to clone AQP1 DNA as described previously [ 27 ] .
